面对习近平于2019年1月2日于北京人民大会堂的《告台湾同胞书》40周年纪念会讲话，强调中国统一大势不可挡，两岸制度差异不会是统一阻碍等说法，蔡英文在当天下午即召开记者会回击。蔡英文强调“一国两制”是台湾绝大多数民意坚决反对的，因此“反对一国两制”是台湾共识。而台湾始终未接受“九二共识”的原因，就是因为九二共识在中国大陆的定义下就是一国两制。借台人对“一国两制”的反感，蔡英文在讲话中将一国两制与九二共识划上等号，借以巩固其“不支持九二共识”的“正当”理由。

蔡英文同时对中国大陆喊话，呼吁中方“必须正视中华民国台湾存在的事实，而不是否定台湾人民共同建立的民主国家体制”，同时“必须尊重2,300万人民对自由民主的坚持，而不是以分化、利诱的方式，介入台湾人民的选择”。
此外，蔡英文还强调，中方必须以和平对等的方式处理双方之间的歧异，而不是用打压、威吓，企图让台湾人屈服，最后更要求中方必须是“政府或政府所授权的公权力机构，坐下来谈”。蔡英文指出，任何没有经过人民授权、监督的政治协商，都不能称作是“民主协商”，暗指“橡皮图章”式的机构无法是为经过人民授权监督，也因此无法与之协商。
至于习近平在相关讲话中所提到的两岸融合与心灵契合，蔡英文则强调心灵契合应建立在彼此的相互尊重与理解，建立在两岸政府务实处理有关人民福祉的问题上。蔡强调，“施压国际企业涂改台湾的名称，不会带来心灵契合；
买走台湾的邦交国，也不会带来心灵契合；军机、军舰的绕台，更不会带来心灵契合”。 对于九合一大选中民进党的大败以及韩国瑜的胜选结果，蔡英文仍强硬表示，“选举结果绝不代表台湾基层的民意要放弃主权，也不代表在台湾主体性上做出退让”。
